BslA mCherry Expression Cassette
BslA mCherry is a composite part that consists of a transcription terminator (parts B0011 and B1002), a P43 constitutive Bacillus promoter, RBS (Bacillus; AGGAGG; 7 bp upstream of start ATG codon), coding regions for the secretion tag LipA and hydrophobin BslA, a GS linker protein (a very linear protein used to link BslA and mCherry, while still allowing for correct folding), and a coding region for mCherry. There are two terminators to ensure that transcription ends. LipA is a secretion tag that helps mark the protein for secretion from Bacillus cells. BslA is a hydrophobin that binds to a variety of hydrogen bonds and is hydrophobic. The mCherry is an RFP or a red fluorescent protein. The BslA linked to mCherry creates a fusion protein that attaches itself to any hydrogen bond and will have a red fluorescent colour that will be detectable to the naked eye and under UV light.
